A global median absolute deviation (MAD) was computed from the gene expression values by taking the median deviation for each set of technical replicates, using either the Ct values or log2 calculated abundances.
The ability to surgically achieve the target alignments was assessed in a previous study by the surgeon authors [ 15], reporting that median deviation for femoral component alignment was ≤ 1.8° in all planes, and median deviation from tibial component alignment was ≤ 1.3° in the frontal and sagittal planes and ≤ 6.0° in the transverse plane.

Each well was converted to a z score using the median and median absolute deviation for each repeat: The minimum of the two scores for each siRNA was selected to represent that siRNA.
The median absolute deviation for a data sample [25] is defined as MAD = median |Yi – median(Yi |), where |Yi| is the absolute value of Yi.
The median standard deviation for each individual estimate is <10%.
The within-group correlation of these ΔCT values was then determined by calculating the median absolute deviation for all the samples within the same experimental group.
