To map the distribution of gene expression levels across the developing cortex, a series of sagittal sections across the latero-medial extent of the cortex were stained for ISH.
Examination of expression patterns for Fgfr3, Cdk6, and Eomes in OSVZ along the entire rostro-caudal and latero-medial extent of the developing cortex revealed distinct domains of high and low levels at multiple locations, outlining complex maps across the early postnatal cerebral cortex (Fig 6).
Apical/basal sectors were split at half the cranio-caudal height of the prostate; anterior/posterior sectors at the level of the urethra; right/left sides by the midline; and lateral sectors were defined as extending 2 cm from the midline or in smaller glands up to the medial highest extent of the anterior horn of the peripheral zone.
The high-frequency region H1 (Figure 7) had a medial-lateral extent of approximately 25 mm, which is slightly more than twice the length of the isofrequency contour seen in the posterior high-frequency region of the macaque.
Fortunately, a preoperative MRI had identified the medial-most extent of tumor as lateral to the wing of the ilium (Fig. 6).
Indeed, a major concern is that PPC inactivations may be only partial, given the large medial-lateral extent of rat PPC (as the paper notes, ∼4mm), the muscimol infusions are likely only partially inactivating PPC, and the lack of effect could be due to compensation by the spared neurons.
Qualitative assessment of transplant size showed that all surviving hGDACNTF transplants were larger than hGDABMP transplants in terms of both their rostral to caudal and lateral to medial extents (Fig. 2 A D).
To ensure that the injury was complete and that the depth was uniformly 1 mm, a 30 gauge needle was again inserted into the transection site to a depth of 1 mm and slowly passed through the medial to lateral extent of the injury.
