We identified separate QTL affecting different measures of virulence: the number of G. mellonella larvae alive at 5 days and 8 days post-inoculation, which may reflect virulence factors that act at different stages of infection.
There was no significant interaction between treatment and parasite genotype on either measures of virulence (change in RBC density: = 2.90, P = 0.58; change in mouse weight: = 3.66, P = 0.45).
Direct tests of the hypothesis should include comparisons of the expression of virulence factors and in vitro measures of virulence between isolates of the same species of the microbiome obtained from healthy/young and ailing/old individuals.
If the microbes are responding to their local environment, it is not clear to me why in vitro, rather than in vivo measures of virulence or gene expression would be that informative.
Examining this relationship, i.e. how host fitness varies with parasite transmission success, has certain advantages because the different measures of virulence among studies are not always easily comparable nor do they necessarily imply the same thing for host or parasite fitness (e.g., host mortality versus weight loss).

Growth, in this species at least, is a good measure of virulence: the nastier the strain, the faster it multiplies.
For HIV-1, the most direct measure of virulence is the time from initial infection to the development of clinical AIDS and death.
Finally, LT50 (the estimated time it takes to kill 50% of the infected fragments) was calculated as a temporal measure of virulence.
In addition to our in vivo measure of virulence, we also measured growth on solid medium so that we could distinguish between QTL with specific effects on virulence from QTL affecting growth both in vitro and in vivo.
In Drosophila, host survival rates can be used as a measure of virulence.
Our definition of suitability required that the murine virulence trend of the clinical isolates be predictably resolved in flies; with host survival being our measure of virulence.
