For unit separation, signals were bandpass filtered (500 5000 Hz) and AP automatically detected and clustered using the wave-clus software57 with standard settings (www.vis.caltech.edu/~rodri/Wave_clus/). Isolated units were separated into regular spiking cells (RS) and interneurons (IN) based on mean firing frequency (RS < 10 spikes/s and IN > 10 spikes/s).
The mean firing frequency of the entire population, measured in each cell immediately after obtaining the cell-attached recording, was 15±15 Hz (n = 76).
The mean firing frequency was calculated from the last 600 ms of each recorded trace where the firing rate reached a steady state (see Fig. S1C for a graphical display of this procedure).
First, analysis of ChR2-mediated glutamatergic responses in MSNs was repeated at near physiological temperatures (32 34°C) at which CINs are spontaneously active (mean firing frequency in cell-attached recordings = 1.6±0.3 Hz, n = 6).
Units from MS/DB with mean firing frequency < 10 Hz were classified as slow-spiking, and with mean firing frequency > 10 Hz as fast-spiking.
They analysed the time that it takes the stimulated neuron pool to rise to its mean firing frequency.

The mean firing frequencies from 6 to 40 weeks are unchanged in the WT, whereas in the ATXN2 Q127 mice over the same age range the distributions progressively shift to slower firing frequency.
SS activity produced by the model was normalized to the mean average firing frequency of the PC in control mice.
The ventral border of the STN was recognized by a decrease of background noise and a decrease of multi-unit activity across a distance of 0.5 to 2 mm, and, passing the ventral border of the STN, a sharp decrease in background firing was found, with more regular firing units (mean fire frequency 20 to 80 Hz) of the substantia nigra (Fig. 1).
Figure 7 summarizes the average firing frequency, regularity of firing and fraction of cells firing regularly as a function of age and genotype.
