The maximal current (iv max and the decrease of ic during the charging of the double layer is measured by a double-beam oscillograph, to show its dependence on applied voltage, supporting electrolyte (pH value), surface-active substances and catalysts of proton reduction.
Currents measured immediately after the final step to −140 mV, were normalized by the maximal current (Imax) and plotted as a function of the preceding membrane potential (Fig. 4A, indicated by a grey arrow).
In contrast to A749G, mutation G407R reduced maximal current amplitudes and caused no change in activation voltage dependence.
Penicillin (5 mM), in contrast, significantly reduced maximal current amplitudes and decreased the concentration required to trigger currents with half maximal amplitudes (Table 1 and Figure 6B).
