The objective of this study is to investigate the role of nanofiber matrix alignment and mechanical stimulation on MSC differentiation, focusing on elucidating the relative contribution of each parameter in guided regeneration of functional connective tissues.
We show that caveolin-1 (Cav1) favors cell elongation in three-dimensional cultures and promotes Rho- and force-dependent contraction, matrix alignment, and microenvironment stiffening through regulation of p190RhoGAP.

To account for both frequency and function, Youn and Simon (2011) (referred to hereinafter as YS) model mutation type, account for sample-specific mutation rates and incorporate BLOSUM80 (BLOcks Substitution Matrix) alignment scores (Henikoff and Henikoff, 1992) into their approach.
