This structure of the homothallic MAT locus, where two MAT idiomorphs are linked but not fused on the same locus, is also present in some other Dothideomycetes (e.g. Cochliobolus species, Stemphylium species) [ 137, 138].
For example, the MAT1-1 ancestor to heterothallic Aspergilli diverged from E. nidulans and its remnant locus organization can be found in A. alliaceus, the only known Aspergillus species for which both MAT idiomorphs are adjacent to one other at a single locus [ 26].
The nucleotide sequence of the MAT idiomorphs was similar among these seven species within Grosmannia: it ranged from 72% between G. clavigera and G. huntii to 97.5% between G. clavigera and L. longiclavatum (Table S2 and Figure 3, A and B).
A multiplex PCR for identifying MAT idiomorph was performed as previously described [ 39].
In a recent study we cloned the MAT genes of eight PAC species [ 44]. and expected the MAT idiomorphs to be similar to those of their helotialean relatives.
MAT1-1 and MAT1-2 idiomorphs were 2294 and 2693 bp in length, respectively, and each contained a single putative open reading frame (ORF) and intron similar to MAT loci of other loculoascomycete fungi.
We further characterized the MAT idiomorph that is responsible for the mating type of the sequenced strain.
In so called pseudohomothallic species, represented in this study by Neurospora tetrasperma, two haploid nuclei of opposite mating-type (mat a and mat A) are maintained in each heterokaryotic ascospore progeny and vegetative cell, while in true homothallic species mat-genes from both idiomorphs are found within a single haploid genome.
Heterothallic (outcrossing) ascomycete fungi carry either one of the two idiomorphs at the MAT locus, and two isolates bearing complementary MAT1 idiomorphs are required to mate.
Within the last few years the sexual (teleomorphic) state was discovered and assigned to the genus Petromyces, and although the fungus is heterothallic, some strains contain both mating-type (MAT) idiomorphs and exist as functionally bisexual- meaning that either idiomorph is capable of outcrossing [ 4].
