In contrast, perfusion of Scn5a+/Δ hearts with 1.0 μ m propranolol significantly increased epicardial APD90 to 72.5 ± 1.2 ms (n = 5, P < 0.05), and significantly shortened endocardial APD90 to 24.5 ±0.3 ms. These marked, contrasting effects of 1 μ m propranolol upon epicardial and endocardial APD90, significantly altered ΔAPD90 to −48.0 ± 1.2 ms (n = 5, P < 0.05).
However, in marked contrast to its effect on K8.1 and ORF59 expression (Figures 1 and 2), 4OHT had no significant inhibitory effect on TPA- or doxycycline-induced vIL6 induction (Figure 7A).
In the first postnatal days, the neurotrophin dependency of primary sensory neurons means that nerve injury causes substantial sensory neuron death (Huang & Reichardt, 2001) but even after this period, for the first 3 weeks of life little or no neuropathic pain results from nerve injury, in marked contrast to the effect in adults (Howard et al., 2005).
