They established a genome-wide map of nucleosome locations and the map shows which region in the genome is occupied by nucleosomes.
Previous studies analyzed the dependence of nucleotide substitution rates in the yeast genome by comparing their positions on a map of nucleosome locations [ 9, 25].
Jiang and Pugh developed a compiled reference map of nucleosome positions in Saccharomyces cerevisiae, using multiple maps independent of our study (Jiang and Pugh 2009a, b).
We chose yeast because we wanted to compare the predictions to a base pair resolution map of nucleosome positions in yeast (Brogaard et al., 2012).
More recently, a complete high-resolution map of nucleosome occupancy across the whole genome of yeast has shown that 81% of the yeast genome is covered by positioned nucleosomes [ 27].

Detailed histone modification profiles on positioned nucleosomes near TSSs revealed interesting patterns, some of which could not be observed previously without the map of nucleosome-resolution modifications.
