The 456 amino acid protein harbors four major functional domains corresponding to its function as an E3 ubiquitin ligase (Table 2) [ Imai et al., 2000; Shimura et al., 2000; Zhang et al., 2000].
Both the longer and shorter isoforms contain the major functional domains of the protein, including the GTPase domain, and are functionally active in biochemical assays [ 29, 36].
Like other members of this family, ERs contain three major functional domains, including an N-terminal domain that harbors a transcriptional activation function (AF-1), a DNA-binding domain, and a C-terminal ligand-binding domain (LBD) hosting a ligand-dependent transcriptional activation function (AF-2).
This chapter defines the major functional domains of neurons and glia, identifies the cellular structures and organelles that comprise those domains, and describes mechanisms required to generate and maintain the functional architecture of these cells.
EBNA1 consists of two major functional domains, a carboxy-terminal DNA binding domain, and an amino-terminal chromosome tethering domain [4], [9].
To provide additional insight into the mechanism of BRCA1 downregulation, we conducted functional mapping studies using expression constructs encoding BRCA1 variants lacking major functional domains (Fig. 5A).

Furthermore, we found that a nonsynonymous mutation on EPAS1, G305S, might be the major contributing factor for the high-altitude hypoxic adaptation, as the codon 305 is not only conserved across all known vertebrates but also located in the PAS-B domain that is the major functional domain binding to its ligands (Key et al. 2009).
Intriguingly, the C-terminal truncation of M2 protein containing only the first 60 amino acids still possesses the ability to upregulate the accumulation of autophagosomes and coimmunoprecipitate with Beclin-1, suggesting that the major functional domain of M2 to interact with Beclin-1 is in the first 60 N-terminal amino acids [ 6, 32, 33].
In mammalian somatic cells, molecular composition of major functional nuclear domains has been studied in detail [ 1].
