A point mutation in the Drosophila gene that codes for the major adult isoform of adenine nuclear translocase (ANT) represents a model for human diseases that are associated with ANT insufficiency [ stress-sensitive B (sesB )].
This is because the CAG repeat, which is transcribed into two different CACNA1A mRNA isoforms by an alternative splicing at the intron46/exon47 splice junction, is placed within a 3′-extended coding sequence of the major mRNA isoform, resulting in the CAG repeat being translated into a polyQ tract [ 1, 15, 65].
Among AS events, 56% were associated with a switch between two functional protein-coding isoforms (N = 9,098, referred to as modified protein), 37% with a switch between a protein-coding and a non-coding isoform (N = 6,067, gain/loss-of function), and 6% with a switch between two non-coding isoforms (N = 1,008, non-coding).
To test if PAG and cortex have different overall expression patterns, we first performed hierarchical clustering using either all initially assembled transcripts or the major isoform for each protein coding gene (Additional file 2: Figure S2).
