Total RNA was purified from 170 dph isolated juvenile sexually differentiating gonads with Trizol reagent (Invitrogen- Live Technologies, Scotland, UK).
Reverse transcription into cDNA was performed using Superscript III RNAse H-reverse transcriptase (Live Technologies, Darmstadt, Germany) with dT15 (TIB-Biomol, Berlin, Germany) and random hexamer primers (Promega, Heidelberg, Germany) as previously described [ 26].
Cells were cultured at 37 °C and 5%% CO2 in Dulbecco's Modified Eagle Medium (DMEM) supplemented with GlutaMAX-1, 10%% fetal bovine serum (FBS), 100 U/mL penicillin and 100 U/mL streptomycin (Gibco Live Technologies, USA).
Cells are trypsinized with trypsin 0.25% (Live technologies, Darmstadt, Germany) for 12-15 min (up to 25 minutes) at 37°C, washed hereafter and seeded onto filters at a seeding density of 4 × 10/well or 2 × 10/ well depending on the time point planned for the experiment.
Briefly, after Ficoll gradient centrifugation (Pharmacia, Uppsala, Sweden), mononuclear cells were collected, washed, and adjusted to 1 × 10 cells/mL in RPMI complete medium (consisting of RPMI 1640 medium (Live Technologies, Paisley, Scotland) supplemented with gentamicin (150 g/mL), L-glutamine (2 mmol/L), and 10% fetal calf serum (all from Gibco-BRL, NY, USA)).
