The predominant safety concern is administration of oxytocin for induction, or more likely, for augmentation of labor.
We observed that glucose and palmitate together appeared to reduce INSIG1 protein (data not shown), which likely contributed to augmentation of nuclear SREBP1 under these conditions.
These data suggest that patients with secondary RLS are less likely to develop augmentation than those with primary RLS.
According to the multivariate analysis in our study, patients with more previous depressive episodes and/or psychotic symptoms were more likely to receive augmentation therapy.
However, pramipexole may be less effective in patients with primary RLS (Ondo et al. 2004) who have previously been treated with other dopaminergic substances since they may be more likely to develop augmentation of the disease (Winkelman & Johnston 2004).

However, SPAK directly affects vascular smooth muscle contraction through WNK1 (52) and WNK3 (53) signalling and NKCC1 (27), the activity of which is necessary for maintaining vascular smooth muscle tone (54), so it is likely that reduced augmentation also reflects hypophosphorylation of NKCC1 in blood vessels in a parallel fashion to our findings in the kidney and other tissues (Figs 2 and 3).
Thus, it is likely that the augmentation of ElvI post-training in our cohort represents a beneficial increase in myocardial performance, primarily caused by improved contractility (as suggested by the increase in S′ velocity along with no change in E′ velocity, a relatively load-independent indicator of ventricular relaxation).
Women with a history of cesarean delivery were more likely to have oxytocin augmentation than those without uterine scarring (41% versus 20%; P=0.037).
Thus, it is likely that the acute augmentation of GSIS at high glucose by GLP-1 is not dependent on elevated substrate oxidation and ATP production but may utilize the Ca2+-mediated and SNAREpin/Munc protein conformational energy dependent fusion of the docked secretory granules with plasma membrane to release their insulin content.
