At a broad level, comprehension processes for narrative content have been studied in the realm of text (McNamara & Magliano, 2009 for review), and readers' eye movements have been shown to differ based on their comprehension (reviewed by Rayner, 1998).
Furthermore, there are numerous widely cited studies that pioneered entirely new areas of research that used a single complex naturalistic stimulus (Hasson, Nir, Levy, Fuhrmann, & Malach, 2004; Yarbus, 1967), a single real-world interaction (Simons & Levin, 1998), or, when studying high level comprehension, of a single narrative (Anderson & Pichert, 1978; Bransford & Johnson, 1972).
