The presence of full-length complements of viral genomic RNA is a hallmark of RNA virus replication within an infected cell.
Because imbalances in the synthesis of genomic RNAs and their full-length complements are common during RNA virus infections, false priming of the more abundant strand during the RT step of the assay is likely to inhibit accurate quantification of the less abundant strand, when standard unmodified virus-specific primers are used for reverse transcription and qPCR.
