Use of the Terminal deoxynucleotidyl Transferase (TdT) end labeling method canceled out fluctuations in labeling efficiency depending on the target sequences caused by in vitro transcription using biotinylated UTP and/or CTP, because the activity of TdT does not depend on the sequence of the target [ 41, 42].
For FCS measurements with fluorescently labeled ligands, fluorescence fluctuations were recorded for two 30 s intervals at a laser power of 0.3 kW/cm following a 10 s prebleaching step at a laser power of 0.2 kW/cm.
