Jagged-1 has been found to be highly expressed in hypertrophic astrocytes within and around active MScl plaques lacking remyelination, while, in contrast, there was negligible jagged-1 expression in remyelinated lesions suggesting involvement of the Notch pathway in remyelination in MScl [21].

This time point correlated with the onset of robust Jagged-1 expression in endothelial cells of vessels directly downstream of the attachment site.
It mediates tumour angiogenesis through various angiogenic factors such as IL-8 and VEGF, and also is a critical regulator of VEGF and jagged-1 expression via a JNK- and AP-1-dependent pathway (Johnston et al, 2009).
These data show that coronary vessels upregulate Jagged-1 after attaching to the aorta to receive arterial blood flow, and that Notch3 is required for pericyte to caSMCs differentiation, possibly in response to Jagged-1 expression at the arterial remodeling zone.
Moreover, transforming growth factor (TGF -ß, a cyTGF -ß upregulated in MS, was able to trigger Jagged-1 expression in reacytokineman astrocytes, which are known to border MS plaqupregulated remyelination, while Notch-1 and the downstreaMSeffector gene Hes5 localised to immature oligodendroglial cells [ 101, 105].
Human and murine MSC have been shown to express toll-like receptors (TLRs) and the ligation of TLR3 and TLR4 by their respective natural ligands, double-stranded RNA and LPS, prevented the MSC from inhibiting T cell responses by the down-regulation of Jagged-1 expression on MSC [ 119- 121].
