Involvement of the cluster in chromomycin A3 biosynthesis was demonstrated by disrupting the cmmWI gene encoding a polyketide reductase involved in side chain reduction.
Biochemical analysis of the NcpA1 and NcpB1 adenylation domains coupled with the recent characterization of the (2S,4S -5-hydroxyleucine dehydrogenase NcpD, which is involved in the biosynthesis of the nonproteinogenic amino acid residue l-4-methylproline from l-leucine, support the involvement of this cluster in nostocyclopeptide biosynthesis.
Functional enrichment suggested strong involvement of this cluster in the Notch signaling pathway (Benjamini corrected P = 4.01 × 10−), which plays a major role in neurogenesis and neuronal differentiation (fig. 6 A).
The involvement of this cluster in neural development was further supported by its significant enrichment in postsynaptic density, the Alzheimer's disease-presenilin pathway and the brain (Benjamini corrected P = 0.003, 0.001, and 0.05 respectively, fig. 6 A).
Cluster 14 contains many genes for proteins known to be involved in the cell cycle (GO term enrichment analysis of this cluster returned P-values of 5.2 × 10-60 for 'cell cycle' and 2.9 × 10-51 for 'mitosis') and supports the involvement of other cluster 14 genes in this pathway.
