The structural analysis reveals the features of the asymmetrically engineered Fc in comparison with symmetric Fc and how each asymmetrically introduced substitution contributes to the improved interaction between asym-mAb23 and FcγRIIIa.
To confirm our hypothesis, we deleted nucleotides −64 to −51 from the pIL22R1 to construct pIL22R1ΔLBS-like (−64 to −51), and introduced substitution mutations in the LBS-like sequence to construct pIL22R1-mLBS-like (Figure 3A).
The established and introduced substitution matrix allows calculating diverged domain swapping experiments and eases the HisKA substitution which may be more challenging than the experiments reported.

As both enzymes exhibit approximately 10-fold slower rates of AMP and Val-tRNAIle formation compared with that of the wt enzyme, the kAMP/ kVal-tRNA ratio, reporting the extent of excess ATP consumption, remains unaffected by the introduced substitutions (Table 1).
