To avoid falling into this trap, for each elongation the model-data error was computed as the sum of the mean squared error during the elongation phase plus a set of seven mean squared errors computed over seven intervals of the relaxation phase (for each time constant in the model, we picked the interval that started with the end of the elongation and lasted three times the time constant).
We evaluated the quality of our proposed methods using these properties in comparison with the conventional approach based on visual detection of relaxation interval limits.
Observations of five myo-3::GFP expressing cells for 30 minutes each showed an average of 0.9±0.3 contractions/min (mean ± SD, n = 5), with each contraction lasting 11.5±1.6 seconds, and intervals of 102±63 seconds of relaxation between contractions.
On the other hand, if we have two extremist groups on both sides of the opinion interval the relaxation is much slower and this is manifested in a much longer relaxation time.
The relatively long inter-train interval was needed due to the markedly slowed rate of relaxation in the presence of 9-AC, as this allowed time for muscle force to return back to baseline prior to the subsequent train.
The latter seems unexpected as the interval of temperatures covers the α-relaxation temperature, which is conventionally associated with activation of additional mechanisms for inelastic flow.
However, over four minutes, unrelaxed force develops due to an extreme elongation of relaxation time so that the muscle does not relax to baseline during the two second intervals between stimulus trains (Fig. 2).
The indenter was displaced toward the top surface of the sample at a constant strain rate by keeping an invariable displacement interval of 0.1 Å and adjusting the relaxation time for each displacement interval.
Recall that the goal of the proposed algorithm is to automatically and accurately determine interval relaxation limits of time point t1 and t2 in Figure 1b.
In this temperature interval, molecular mobility was characterized using different definitions of the relaxation times and calculating a distribution of relaxation times in the system by studying the motions of small regions of the lattice.
