TopoIIα's ability to cleave DNA in a reversible manner makes it an ideal target for agents such as doxorubicin and etoposide, which poison the enzyme via the trapping of the transient reaction intermediate composed of TopoIIα bound covalently to the 5′ end of the cleaved DNA strands (cleavable complexes), preventing religation of DNA [ 43].
