The band at 26 kDa is consistent with cleaved GST, which, together with several other intermediate molecular-mass bands detected by the GST antibody, are the probable result of some degradation occurring through protein expression and purification.
Subtilisins are further classified into six subfamilies namely, true subtilisins, high-alkaline proteases, intracellular proteases [ 4], phylogenetically intermediate subtilisins (PISs) [ 7, 8], high molecular mass subtilisins (HMSs) [ 9, 10], and oxidant stable proteases (OSPs) [ 11, 12].
Using this approach, we were able to identify the molecular mass of ten different intermediates produced during SBP-H2O2-mediated degradation of Trypan Blue.
The sequences and parameter details of the transcripts were sorted according to gene families and compiled in Additional file 2. The high-level expression of a core-glycosylated ABCB6 with a correct molecular mass, the specific Mg2+-dependent ATP binding and the formation of a catalytic intermediate strongly argue against this possibility.
It is, however, also possible that no specific transport proteins are required since yeast peroxisomal membranes contain pore-forming proteins that allow passage of small molecules with a molecular mass up to 400 Da, which is sufficient to allow passage of these intermediates (Antonenkov et al. 2009).
