Since patients on warfarin are on multiple medications, which may all potentially interact with warfarin, sometimes with opposite effects, we evaluated the sum of these interactions which explained up to 2.2%% of the variance, showing that much greater effect of co-medications can be determined by taking into account the effect of all of them.
In conclusion, we report 26 replicated variance eQTL and 57 replicated cis epistatic interactions, which explain up to 16% of the variance of our phenotypes.
We finally reached a model containing VKORC1, CYP2C9*2 and *3, PROC, EPHX1, GGCX, ORM1-2, age, bodyweight and drug interactions which explained 73% of the variance in warfarin dose (Table 4).
As seen in our structure, an alanine or valine substitution would completely abolish the interactions with the 2-NH2 group and the equatorial OP1 of cGMP, whereas a serine substitution would affect only the latter interaction, which explains the changes in cGMP affinity observed with each mutant.
