To interrogate the loading of cohesin onto chromatin, we quantified the signal intensity of extraction-resistant SCC1 staining in fixed interphase nuclei (Schmitz et al, 2007).
Although depletion of the cohesin-loading factor SCC4 (Ciosk et al, 2000; Watrin et al, 2006) reduced the intensity of extraction-resistant SCC1, depletion of the splicing proteins NHP2L1, MFAP1 and SART1 had no discernible effect (Fig 4B).
