Mikhaylova and Nurminsky [ 32] took advantage of the spermatogenesis timeline and obtained RNA from the first wave of germline differentiation by dissecting larval testes collected from days 4 to 7 (second-instar larvae and the point in which the third-instar start to pupate, respectively).
These differences are established during embryogenesis [5], [7], when hatched first instar nymphs already display variation in which segments become enlarged as well as in the magnitude of their size increase (Fig. 1C).
Have they tested these in 3rd instar central larval brain neuroblasts, in which the transient Pros expression was shown to induce neuroblast quiescence?
We previously used a nutrient restriction (NR) protocol in which third instar larvae are transferred from a yeast/glucose/cornmeal to an agarose medium (Cheng et al., 2011).
The only published reports we can find where C. ulrichii has attacked other species than moose, are a case in which first instar larvae were found in the conjunctival sac in the eye of a human [ 19], and another case where 39 young larvae were deposited by a female C. ulrichii on the upper lip of a human [ 1].
The larval period was defined as the interval in which third instar larvae stop feeding, purge their gut and wander toward a suitable pupation location, early pupation was defined as 0 3 days after initiation of pupation and mid-pupation was defined as 4 6 days after initiation of pupation.
Encarsia embryos and larvae are in intimate, direct contact with the host's hemolymph throughout their development, whereas Eretmocerus become in contact with the host's hemolymph only in the third instar, due to the unique capsule in which the larval wasps reside [39].
To check whether Abd-B is permanently required to repress CCAP/Bursα expression, we knocked down Abd-B in first instar larvae (elav-Gal4 UAS-Abd-B-RNAi UAS-dicer2; although this driver is expressed in the embryo, RNAi is not effective until the larval stages) and, in second instar larvae we observed several cases in which ENs of segments A5 7 expressed neuropeptides prematurely (Fig. 5C).
Application of JHA also did not increase the length of the 4th instar (Table 3), but in all experiments, except those in which JHA was applied within 20 hours into the 4th instar, 5th instar larvae molted into a 6th supernumerary instar (Fig. 4).
Manduca spp. behavior, predation, and growth assays were conducted with first- and second-instar larvae, except infestations M1 2010 and M4 in 2011, in which larvae were reared from the first through fifth instars on plants; and egg predation assays, in which M. sexta eggs were used.
