Alternative large insert approaches, like fosmid di-tag sequencing [ 20], have been documented to suffer from low library complexity, which may be overcome by using larger amounts of input material, but they have an additional disadvantage as they are restricted to a fixed insert size of approximately 40 kb [ 16, 20, 35, 36].
