A real-time RT-PCR analysis using primer sets designed to amplify from exons 3 to 4, exons 4 to 5, exons 5 to 6, exons 6 to 7, and exons 7 to 8 of the mouse ER-α gene revealed the existence of alternatively spliced ER-α mRNA and its putative transcription initiation site, located between exon 4 and exon 5.
Each clone corresponds to a different initiation site located at positions 28, 55 and 80 (Fig 4b, Fig. 3c).
This indicates the larger transcript was likely transcribed from a different 5' initiation site located upstream of the gene.
The confirmed initiation sites are shown in bold in Fig. 2. The 3'-most initiation site, located 16 nucleotides upstream of the 5'-end of the previously reported cIRK1 cDNA, was numbered +1.
RACE analysis on mRNA extracted from primary skin fibroblast detected a single transcription initiation site located 14 bp upstream of the ATG initiation codon, while the 3′ untranslated region (UTR) extends to 501 nucleotides downstream of the termination signal, and contains a non-canonical AGTAAA polyadenylation signal (Fig. 2B).
When we superfused the CPVT multicellular preparations, we observed that KN-93 stabilized calcium activation, reverting to the typical pattern observed in normal tissue, characterized by a single initiation site located on the periphery of the preparation where the source-to-sink load was favorable.
For example, the mouse translation initiation site is located 120 nucleotides upstream of the predicted human start codon.
Thus, the P3 RNA initiation site was located at approximately nt 160, and the D1 site was confirmed to be at nt 300.
The transcription initiation site is located inside the conserved box 6 8 bases before 3'-end.
In Arabidopsis the polar initiation site is located towards the basal end of epidermal cells.
