On the basis of multiple three-dimensional structures of SufS, the shortness and decreased flexibility of the active site loop containing the catalytic Cys364 likely explain the catalytic inefficiency of group II desulfurase enzymes compared to group I enzymes, which have a flexible catalytic cysteine loop that is 11 amino acids longer than the group II enzymes.
