The biotin-free α subunit and the biotin-containing α subunit showed a REES of +6.9 and +5 nm, respectively, indicating a restricted mobility of tryptophan surrounding solvent molecules in these proteins (Fig. 4, a and b).
The origin of this radical is also an anionic semiquinone indicating a higher mobility of the FAD due to a smaller linewidth.
In contrast, for the D2O solution of the random copolymer, weak signals of methyl groups on cholic acid moieties were observed, indicating a greater mobility of the cholic acid-containing segments in this case.
We observed, as in the case of cytoplasmic FLIP, that nuclear photobleaching (marked with dark blue box) leads to a corresponding 40% decrease in the intensity of cytoplasmic GFP-FTO signal (marked with red box), and also an almost 80% drop of GFP intensity in the nuclear region adjacent to the photobleached region (marked with light blue box), indicating a high mobility of nuclear GFP-FTO.

These results indicate a reduced mobility of the SUMOylation-defective mutant CBP in the nucleoplasm, and suggest that SUMO modification regulates the mobility of CBP in the nucleoplasm, perhaps by modifying its association with nuclear proteins, chromatin or macromolecular structures such as nuclear bodies.
