Using a monomerised RadA as our starting point, we have adopted two parallel and mutually instructive approaches to mimic the human enzyme: firstly by mutating RadA to increase sequence identity with RAD51 in the BRC repeat binding sites, and secondly by generating a chimeric archaeal human protein.
Finally, to improve success of recombination of genes with low level of identity, one can also increase sequence identity between two genes.
Taken together these results support a process of gene conversion rather than gene convergence, as the major cause for increased sequence identity between paralogues.
In addition to again showing a role for common polymorphisms in marking differences in local methylation, these analyses of sibling pairs highlighted an additional component of heritability most easily explained by the increased sequence identity in IBD regions.
