The trends are consistent with two observations from the digestibility data above: (1) the overall glucose yields from wood chips increased with increasing pretreatment time and (2) the 4-min pretreated chip exhibited digestibility spatial effects across its thickness, whereas spatial effects were not observed for chips pretreated for longer times.
Increasing pretreatment time only seems to have minimal influence.
As expected, the recovery of solids decreased with increasing pretreatment time (Fig. 2c).

For Stage 2, glucan plus xylan yields reached peak values of 64.2 and 61.9% after 28 and 11.4 minutes for pretreatment at 180°C and 200°C, respectively, but sugar yields continually dropped with increasing pretreatment times at 220°C for the range of times tested.

In summary, with Accellerase 1000 alone, pH adjustment or extended residence time caused a similar enhancement of monomeric Glc yields (that is, from 46% up to 58% to 59%), but when using an optimized commercial cocktail, pH adjustment caused a bigger enhancement than increased pretreatment time (that is, from 66% to 74%).
Higher glucose and xylose yields were found with increasing pretreatment residence times, especially for the control substrates.
Similar glucose yield improvements were found with the control sample of 33B51 by increasing pretreatment residence times; glucose yield increased from approximately 57% at 5 minutes to approximately 64% at 10 minutes and to approximately 72% at 20 minutes.
