In our preliminary experiments, although we carried out the ligation reactions with same overhang sequences repeatedly in one assemble, we could not isolate our designed plasmids harbouring multiple DNA fragments; by skipping internal DNA fragments, many plasmids included less DNA segments than those that we desired.
