Measurement with the interferometers with phase stepping (done by shifting the G2 grating horizontally across the beam) provides a projection image of phase-shifts.
The simplicity of the design makes the condenser a user-friendly, dedicated device delivering high-contrast quasi-3D images of phase objects.
Imaging of phase objects is especially important for biological systems [27, 41].

Images of phase contrast microscopy show indeed a reduced density of the cell layers treated with either SLN− and SLN+ compared to untreated or sorafenib-treated cells with no alteration of the cell morphology (Additional file 1: Figure SI11).
However, this is unlikely here because BicDPA66 and BicDnull (t = 3) embryos have nearly identical tracking phenotypes in phase II (t-test, p>0.75 for both directions; Fig. 6H, I), and confocal images of phase II microtubules in BicDnull (t = 3) embryos generally appeared fine (Fig. 2E).
For the interpretation of the FOI examinations, the images of phase 2 with the peak ICG enhancement at the hands and fingers were used for the scoring [ 14].
In order to improve the SNR and the image quality of phase retardation images, we employed a custom algorithm of complex moving averaging for Jones matrices.
The image quality of phase retardation is important to visualize clinical data measured by PS-OCT.
