The segmentation method that was used in the present study for identifying the nuclei has been previously developed by our group for other similar applications and a detailed account the method may be found in [ 21].
Much of the research on cell reproduction came together in August Weismann's theory of heredity: he identified the nucleus (in particular chromosomes) as the hereditary material, proposed the distinction between somatic cells and germ cells (arguing that chromosome number must be halved for germ cells, a precursor to the concept of meiosis), and adopted Hugo de Vries's theory of pangenes.
For each cell, the DAPI channel (Fig. 3B) was used to identify the nucleus, and the remaining field was computationally segmented to determine the cytoplasmic compartment for each cell (Fig. 3B C).
For intracellular dye localization, cells were first labeled with VT680, fixed with Cytofix/Cytoperm™ kit (BD Biosciences, USA), then labeled with PI (propidium iodide) (1/1000) to identify the nucleus.
Tumors were counterstained for DNA with DAPI to identify the nucleus of the cells.
The program consists of image-analysis procedures that sequentially segment the individual cells, track them in time, identify the nucleus, and measure the p53 fluorescence intensity in the nucleus and cytoplasm, respectively.
Taken together, these studies identify the nucleus as a novel cellular organelle involved in ALS pathogenesis and open new avenues for the potential development of therapies targeting this fundamental cellular structure.

In summary, we have studied the array nucleation phase and identified the nuclei of both hut species, determined their atomic structure, and observed the moment of appearance of the first generation of the nuclei on WL.
Finally, the films were stained with 0.5 µg ml−1 4', 6-diamidino-2-phenylindole (DAPI) in PBS to identify the nuclei.
