Each of these mechanisms has been studied in detail, leading to the identification of checkpoints that could be defective in autoimmunity (Fig. 2) and thus represent targets for immunotherapeutic intervention.
Identification of spindle checkpoint genes independently shown to be synthetic lethal with SPB damage (Figure 1D), as well as identification of Spc110 transcriptional regulator HCM1, demonstrate the effectiveness of the SGA screen.
