The objective of the experiments was to investigate the outcome of low- and hyper-velocity impacts on two identical target satellites.
But when expressed in combination with SpdCas9 and a pair of sgRNAs specific to two proximal binding sites in HBB, CjCas9 cleaved the HBB target efficiently (32% indels) without cleaving the identical target in HBD (Fig. 8b).
The previous experiments completed in late 2005 aimed to compare low- and hypervelocity impacts on identical target micro-satellites, whereas the new tests used larger micro-satellites as targets and aimed to investigate the effects of impact directions.
One monobody contains the Tyr/Ser binary-code interface (termed YS) and the other contains an expanded amino acid diversity interface (YSX), but both bind to an identical target, maltose-binding protein.
Conversely, CjCas9 was able to selectively cleave the HBD target fairly efficiently (14% indels) without cleaving the identical target in HBB when it was aided by SpdCas9 and a pair of sgRNAs specific to two proximal binding sites in HBD (Fig. 8b).
