Similar neoantigen-targeted vaccine approaches were previously tested6,7 in early-phase clinical trials for melanoma, which, unlike glioblastoma, is a type of cancer that has a high level of mutations and neoantigens.
In fibroblasts, complex I deficiency was secondary to a high level of mutations (68%) (Fig. 1 D ).
Analysis of six genes revealed a high level of mutations with a density of about one per 60 kb.
This observation suggests that the two chosen DNA fragments (hspA and glmM) correspond to domains with a high level of mutations and encode proteins with few functional constraints.
All of these methods suffer from a relatively high level of mutations in the amplified DNA (error rate 3x10-5[ 3x10-5[ 13]ghly non-uniform andlification due to thighly fidelity and low processivity of the Taq polymerase, respectively.
We next followed the mutation rates of these high-efficiency mutated shoots in all the regenerated transgenic tomato plants with mutations, and found a high level of mutation (30 90%) in T0 plants of both Micro-Tom and Ailsa Craig (Fig. 2a).
The current data makes the possibility that such plasmids have a high level of mutation worthy of further study.
Polyploids, such as hexaploid bread wheat, tetraploid durum wheat, and hexaploid cultivated oat, are easily amenable to TILLING probably because genome buffering enables plants to survive a high level of mutation [ 12- 17, 17].
In contrast, a high level of mutation (approximately 40%) is observed with bi-allelic mutation of CEBPA, conferring a better prognosis than CEBPA mutation with wild-type GATA2 (Greif et al, 2012; Fasan et al, 2013; Green et al, 2013; Grossmann et al, 2013; Shiba et al, 2014).
This is unexpected as intergenic regions such as the STAR loci, which consist of repetitive elements dispersed throughout the genome, would be expected to show a high level of mutation and hence evolve at a higher rate than the conserved functional MLST loci where mutations are observed at a very low rate [ 19].
Although this very high level of mutation clustering could, at first sight, be interpreted as evidence for positive selection, we found that silent substitutions were also frequently recurrent (28 recurrent variants in 13 mtDNA sites), with no substantial difference in recurrence rates between silent and missense mutations (p = 0.19; Figure 5 figure supplement 1).
