To determine whether the high frequency, strain-dependent binding of gp41 MPER in IgMhi B cell subsets was mediated by membrane IgM or some other strain-specific molecule, we activated splenocytes from naïve BALB/c and C57BL/6 mice in vitro with BAFF+LPS and measured the fraction of total (IgM+IgG) ELISpots that bound gp41-MPER peptide (Fig. 3).
