Levy et al. [26] reported the effective use of a randomly mutated BoNT/A heavy chain library, displayed on the surface of yeast cells, to identify single amino acids important in the epitopes of anti-BoNT/A monoclonal antibodies.
For the heavy metal library (TTS), cells were grown to mid-log phase in Neff medium [ 30], and separate equal aliquots were treated with 11 μM CdCl2 or 500 μM CuSO4 for 1 h.
The selected variable antibody domain genes of the shuffled human light chains were cloned as ApaLI and NotI fragments in pHenIX containing a human variable heavy chain library (8 × 10; R. Finnern, unpublished data) and introduced into E. coli TG1.
Compared with the gDNA heavy chain libraries, the corresponding cDNA heavy chain libraries had significantly higher percentages of clones using IGHV1 and IGHV3 lineages (Fig. 1), and were biased to certain VH1 and VH3 subfamilies, including IGHV1 18, 1 2 and 1 69, and IGHV 3 11, 3 21, 3 23, 3 30, 3 33, 3 49, 3 7 and 3 74 (Fig. 2).
This update improves the speed with which large libraries are imported and fixes many of the heavy batch issues that cropped up in the first release.
However, the rich medium and heavy metal libraries displayed higher redundancy, with over 53% and 40%, respectively, of ESTs corresponding to genes that were hit by 100 or more total ESTs (from any condition).
Among the four gDNA heavy chain libraries, NIgH and pt1gH showed a similar pattern of various IGHV lineage usage, whereas pt2gH and pt3gH were significantly different from NIgH and pt1gH in using IGHV1, IGHV2 and IGHV6 lineages (Figs 1 and 2).
All five metallothionein genes [ 47] are among the ESTs found predominantly in the heavy metal cDNA library, as are other genes with potential roles in response to oxidative stress (e.g. thioredoxins, glutathione S-transferases and peroxidases).
Previously we had generated antigen-driven variable heavy chain cDNA libraries from sentinel lymph nodes of breast cancer patients [ 12].
Nicola Morgan, who chairs the Society of Authors' children's writers and illustrators group, warned that the report was undermined by its failure to address the heavy losses sustained by library services squeezed by steep reductions in local authority funding.
