At 37°C, fluorescence spectrometry on dilute solutions of the purified protein yields a half-maturation time of 2.8 h, which makes the protein suitable for most cell biological imaging applications (Figure 2A).
Compared to an enhanced GFP variant (eGFP), mCherry matures quickly at 37 °C with maturation half time of 15 min (Shaner et al. 2004).
The other half of this maturation equation is that parents should remain in ignorance of their adolescents' exploits – none of this "add me on Facebook" so I can see photos of you bladdered.
RFP639 is approximately three times more photostable but has a longer maturation half time (1.5 vs. 0.25 h for mCherry).
This protein has spectral properties very similar to Katushka, but its maturation (half time of 75 min) and pH stability are inferior, although the brightness level is comparable to that of mCherry.
It was also noted that in large non-domestic cats, the ossicles become visible radiographically at approximately one year of age or by the last half of skeletal maturation [ 11].
What is interesting is that the content of immature N-glycans diminishes during lymphocyte maturation; half of the N-glycans on CD45 on thymocytes are high-mannose or hybrid-type, while the majority of N-glycans on CD45 on peripheral T cells are ST6Gal1-modified complex-type structures [ 30].
This intensity profile indicates that the half-time of maturation at 25°C is approximately 40 min similar to the time observed for half-maximal increase of Cse4-tdEos fluorescence in a population of asynchronously growing cells.
They propose that the secretion of beta-amyloid peptide Abeta40 and Abeta42 is increased following the overexpression of APBB3, but there are no visible changes of half-life and maturation of APP, or the secretion of secreted APP [612].
A single exponential fit gave directly the maturation half-time <Tβ> = ln(2)/β = 18.5+/−0.4 min, with low cell-to-cell variability, as expected (standard-deviation/mean or coefficient of variation (COV) = 0.16), Fig. 2d.
For comparison, the red FP mCherry has a maturation half-time of 40 min in the same assay (4).
