Grooming was manually scored from the first 10 minutes of the open-field test, and the latency to the first grooming event, the number of grooming bouts and the total grooming time were measured (Fig. 5C).
Fluoxetine restored the total grooming time to the unstressed level in UCMS mice who received a regular diet, but this was not observed in the mice with the high fat diet (RD: T = 0.0, p<.05; HFD: T = 24, ns) suggesting that a high fat diet prevents the reversal effect of fluoxetine on repeated UCMS-induced grooming disturbance.
In the Open Field test, a measure of exploratory behavior and general anxiety, we detected no difference between the deletion mice and WT littermates in total distance traveled, total time of rearing and grooming, time spent in the inner square, and frequencies of urination and defecation (data not shown).
The anti-depression effects were evaluated based on a measured index, which consisted of visiting counts, start latency, rearing number and grooming time.
