Since no tertiary structure information about Noc4p-homologues or related proteins is available we subdivided the Noc4p-primary structure into several subdomains by other means: the different grade of conservation within distinct parts of Noc4p coding sequences from several eukaryotic species served to classify the polypeptide into eight domains (Fig. 2A)(see also Fig. 1 [28]).
We found an overall high grade of conservation on molecular and cellular mechanisms associated with tissues, diseases and aging that is consistent with previous results [ 8, 52, 53].
For example, in the case of the organism we study in this work (zebrafish), a simple analysis of two classic gene families known to have a very high grade of conservation among their members (homeobox and histone gene families) reveals that, at nucleotide level, values of sequence identity for each pair comparison were up to 80% (e.g., typically in the range of 71 80% in the case of homeobox).

Positions other than Q123 and Q126 (having conservation grades of 6 and 7, respectively) display variability amongst the ferritin members (Figure 3 and Figure 6A).
For this purpose, the multiple sequence alignment output of the entire KLK homologous amino acid sequences analyzed in this study was used as input to the program to project the conservation grades of residues on the known three-dimensional structure of the human proKLK6 (PDB ID: 1gvl) [34].
