The role of intrinsic DNA sequences either in in vitro or in vivo nucleosome positioning has recently been at the centre of a debate about the existence of a genomic code for nucleosome positioning.

It would not be surprising if in certain cases sequences coevolved with nucleosome positioning, but there would remain a counter-evolutionary pressure to allow nucleosomes to package all DNA, and so the extent, or strength, of any such genomic code for nucleosome positioning cannot be absolute, that is, the wells of the binding landscape cannot vary too extensively across sequences.

It has been suggested that evolution has harnessed the sequence preferences of nucleosome formation to define a "genomic code" for nucleosome positioning, but the idea remains highly controversial, and it seems clear that other facets of chromosome organization also impact nucleosome positioning (see below).

Although the role of genomic sequence on nucleosome distribution is still a controversial topic, it has been confirmed, both in vivo and in vitro, that DNA sequence imparts nucleosome distribution and there actually exists genomic code for nucleosome positioning [ 38- 40].

The genomic code for nucleosome positioning could be even more diffuse than previously thought, as the structure of the histone core particle reveals that tetranucleotides or possibly even longer sequences are involved, rather than the dinucleotide patterns that were recently discovered [ 4].
