To generate the lymphokine-activated killer (LAK) cells, isolated PBMC and NK cells were stimulated with 6,000 U/mL of human IL-2 (MACS; Miltenyi Biotec, Auburn, CA) for 24 hours.
Common early progenitors in the thymus generate the diverse T-cell subsets: CD4, CD8, CD4 T regulatory (Treg), invariant natural killer T cells and γδ T cells.

Furthermore, at certain time points in the treatment cycles the capacity to generate lymphokine-activated killer (LAK) cells from the peripheral blood mononuclear cells of these patients was significantly augmented above that observed prior to treatment.
Activation with high doses of IL-2 for 7 days is the standard procedure to generate lymphokine-activated killer (LAK) cells, which mediate potent killing activity.
In this line, several protocols have been established to generate mature natural killer cells (NK) from human iPSCs or ESCs, commonly using the hematopoietic cytokines SCF, Flt3-L, IL-3, IL-7, and IL-15 (Woll et al, 2005; Sturgeon et al, 2014; Ferrell et al, 2015) (see also Table 1).
