"game up" is not a grammatically correct phrase.
It should be "game over" or "game up." As for when to use it, it is typically used at the end of a game or competition to indicate that it is finished or that one side has won. An example sentence could be, "After the final whistle blew, it was clear that the game was up for the losing team."
