The cDNA clone of HCT was PCR-amplified and inserted in the pEU plasmid vector (CellFree Sciences, Japan).
For translation, the mRNA of each construct (10 μl) mixed with the same amount of the WGE WEPRO 7240G (CellFree Sciences) and 0.1 mg of creatine kinase to make the bottom layer, was incubated with the 1× SUB-A MIX SGC (upper layer), which includes the amino acid mixture (CellFree Sciences).
Cell-free protein synthesis used the reagents of an ENDEXT Wheat Germ Expression S Kit according to the manufacturer's instructions (CellFree Sciences Co., Ltd ., the bilayer translation method, and a robotic synthesizer (GenDecorder 1000; CellFree Sciences).
After in vitro transcription from the recombinant plasmid, HCT-fused to a C-terminal FLAG peptide (HCT-FLAG) was in vitro synthesized in the wheat germ cocktail (CellFree Sciences, Japan) according to the manufacturer's instructions.
DNA (2 μg) of each construct was transcribed using SP6 RNA polymerase, 25 mM NTP mix, RNase inhibitor and 5× transcription buffer (CellFree Sciences) for 6 h at 37°C without shaking.
The translation mixture that formed the bottom layer consisted of 60 A260 units of the wheat germ extract (Cell Free Sciences Yokohama, Japan) and 2 μg creatine kinase (Roche Diagnostics K. K., Tokyo, Japan) in 25 μl of SUB-AMIX® (CellFree Sciences).
