Interestingly, the band was decreased in the presence of Atxn1 especially by mutant form expression. Expression levels of Atxn1-33Q-DsRed or Atxn1-86Q-DsRed were almost equivalent (Fig 6B).
ΔHS1 deletes a 340 bp region from intron 1 of Zfp386 which might result in different splice forms, impaired expression levels, or expression patterns of this gene.
It is interesting to note that while PNPLA6 and -7 are a pair forming one patatin subfamily, and PNPLA8 and -9 form another (cf. Figure 4), expression levels of PNPLA6 and -9 are high in brain, and those of PNPLA7 and -8 in skeletal muscle.
To this aim, we first assessed the expression of microtubule-associated protein light chain 3 (LC3) after conversion from LC3-I form to its autophagosome membrane-associated lipidated, activated LC3-II form, and the expression levels of p62, a bona fide autophagic substrate.
With these constructs, 11 BVMOs could be obtained in soluble form with good expression levels, using E. coli TOP10 as host (Table 1).
