We have studied a series of structural descriptors intended for describing protein shapes (the radius of gyration, the radius of cross-section, and the coefficient of compactness) and their possible connection with folding behavior, either rates of folding or the emergence of folding intermediates, and compared them with classical descriptors, protein chain length and contact order.
There is enormous diversity in the protein folding behavior from small proteins usually folding with simple two-state kinetics to large proteins usually folding with multi-state kinetics.
To explore the effect of a PMF barrier on the folding behavior of proteins, we modified Zwanzig's model for protein folding with an uphill landscape of PMF for the formation of transition states.
The significantly different stability of their highly similar native folds after transfer into the gas phase, and their strikingly different folding behavior in the gas phase, can be rationalized on the basis of electrostatic interactions such as salt bridges.
Regardless of the exact magnitude of the barrier, our smFS experiments demonstrate that under mechanical force gpW folds and unfolds via a two-state mechanism in contrast with its downhill folding behavior in the absence of force.
We also characterized the folding behavior of the intermediate using stopped-flow fluorescence, protein engineering, and native-state hydrogen exchange.

It is also found that their folding behaviors are very similar to other three-helix bundles.
We predict different folding behaviors for different SAM-I riboswitch sequences.
As growing numbers of assays are developed for enzymatic or folding behaviors of single macromolecules, the range of applications for fluctuation analysis increases.
