Knockdown of an autophagy related gene showed only increased autophagic flux blocking and little change in cell cycle related gene expression (Fig. 4).
Our simulations revealed a flux block in the conversion of methionine to cysteine for MMA, propionic acidemia and cystathionuria (Fig. 3).
Considering these results and taken together with the finding that chloroquine did not increase in LC3-II in the optic nerve after IOP elevation, it is possible that autophagic flux blocking may be involved in these processes.
On the other hand, in the optic nerve, no difference in LC3-II levels in the presence and absence of chloroquine was observed in the glaucoma group compared with the control group (Supplementary Figure S1), suggesting that there was autophagic flux blocking in the optic nerve after IOP elevation.
