On the basis of visual inspection of the Lineweaver Burk plots and the accuracy of fits, compound 3 appears to be competitive with the substrate BAEE, with a Ki of 100 ± 30 μM, and noncompetitive with respect to calcium (Ki = 500 ± 65 μM) (Supplementary Figure S5C and D, respectively; Supplementary Tables S2 and 3).
Figure 2A and 2B illustrate the aggregation curves of compounds 3 and 4, respectively and figure 3A illustrates the IC50 curve fitting of compounds of type I. Ibuprofen was used as a reference for the inhibition of platelet aggregation (Figure 3A) and as expected, showed an inhibition of platelet aggregation with an IC50 of 0.5 ± 0.11 μM (n = 3).
Given this, it was not possible to fit individual compound concentrations as for the polar compound data, and so we used a different approach for data analysis.
